Sanger sequencing

Sanger sequencing can be used to determine the sequence of PCR products or vectors, obtaining yields of up to 1000 bp. Indicated for:

  • PCR products, vectors (DNA cloned in plasmid, phage, BAC ...).
  • Sequence simple genes.
  • Cost-effective sequencing of individual samples.
  • Verification of directed mutagenesis or presence of cloned inserts.

We use BigDye Terminator chemistry v.3.1 (Applied Byosistems) and AbiPrism technology (Thermo Fisher Scientific)

* General recommendation: it is recommended to mix 10 μL of purified DNA (PCR products> 20 ng / μL, plasmids 100-500 ng / μL) and 3 μL of primer (10 pmol / μL). Consult us in case of doubt at the time of preparing the samples.

In case of sequencing failures due to the characteristics of the sample (low quality of DNA or quantity, poorly purified or inadequate primer) the services will be billed as well.

Sending samples

Samples must be correctly labeled and identified. It is recommended to use well sealed 0.2 mL tubes. The PCR or DNA products to be sequenced must be purified and quantified (see if you need these services), and the samples must be diluted in nuclease-free water, avoiding elution buffers from commercial kits.
