BIOGENETICS as part of the researching activities developed about new analytic technologies for nucleic acids, has participated from the beginning in the validation and testing of the new sequencing plattflom based in “single-molecule” MinION from Oxford Nanopore Technologies.
Oxford Nanopore Technologies is the developer of a disruptive sequencing technology that confirms us in the idea, for which we decided to enrol us in the MAP program, about that the future of the sequencing is nanopore based.
Since that the past year Oxforf Nanopore Tech introduced the first world sequencer about the size of a USB memory, showing a new horizon of mobile sequencing, the platform in constant improvement process and evolution, both in terms of hardware as well as software and reagents, offers currently a medium accuracy about 85%, being possible, in specific cases, reach even with the current chemistry and basecalling algorithms a greater accuracy.
For those unfamiliarised with the tecniques of sequencing think that MinION is albe to sequence samples directly (without pre-processing and in “real-time”) it should be clarified that being a really amazing device isn’t capable of this type of miracles, “yet”. And therefore it requires obviously of the extraction, purification and library construction thoroughly; even with DNA ready to be charged in the MinION to speak about “real time”sequencing can lead again to misunderstandings to those outside the genomic techniques. Because during the run is only possible to see in real time, the number of reads and their length, being the basecalling (determination of bases from the signal) and quality determination object of a later “on-line” processing through Metrichor.
Metrichor is based equally in a new concept, thought to maximize the capabilities of mobility of MinION in a collaborative broad range, being able to the taxonomic identification in real time to basecalling, comparing directly the results with reference data bases as for example in the WIMP (What´s in my pot) workflows.
Certainly MinION offers a smaller number of reads (throghput) than other plattforms, even though the performance (Mb/run) is compensated by the length of reads that is capable of providing. Feature that offers also interesting opportunities for several applications focused to food safety based on molecular traceability (authentication-fingerprinting, quality assessment-qualification y foodborne diseases control) for which have long readings (highly informative and segregative) is crucial.
The current massive sequencing (paired end) is normally developed with short reads, due to, between another factors to the novelty and greater difficulty of the available long reads workflows, only available for certain platform also. On the contrary the great length of the MiinION’s reads permit in a easy and quick way the metagenomic taxonomic identification to specie level even in cryptic or complex cases (breeds, high backcrossing, etc…) and even qualities (based on QTLs, candidate genes…).
